Kirkman regretted cutting off ricks hand so early, so perhaps that could be an arguement for someone like Negan to do it, show he means business.
Negan meant Rick's right hand man not the hand itself (However he might still cut off Rick's hand, Who knows?).
His speech and the blanket all indicated that he had killed Daryl ( Fuck!!) but my money's on either Abraham or Glenn since Daryl's death would be catastrophic for the show.
