The bite gives the victims a very strong virus that slowly kills them, because the bacteria that's on the walker's teeths comes in contact with a fresh wound.
My understanding of the whole zombie bite thing, was that the infection we all have weakens our immune system. When a walker bites you all the nastiness in their mouth goes into your blood and with our weakened immune systems we die.
Just look at the flu from (I think) season 4 of the show. It would have been pretty bad to begin with but the weakened immune systems made it much worse.
Possibly true, but then if you think about all the crap they go through: drinking unclean water, eating mouldy food and sleeping in rough conditions, you would think that they would have all popped it by now.
I always thought that infected walkers produce a toxin or something through the salivary glands which merely kills a potential host, possibly by shutting down signals in the brain. The dormant virus is then able to take over.
If the nasty stuff in their mouth causes to kill people with weakened immune systems, how do you explain someone turning after getting bit by someone else who turned very recently.

It's not just some disease from rotting teeth. That doesn't make sense, freshly turned walkers' bites also kill people and bite victims can't even be saved with antibiotics or strong alcohol solutions.

It then revives the brain stem and the parts of the hypothalamus responsible for hunger, as well as starting to spread a strong toxin through the host's body fluids.
Honestly though, it's because the plot demands it. What I said doesn't make any sense from a biology standpoint. It would obviously never work in a real life scenario. But that's the best I can come up with for the TWD universe.

From what I understand a walker bite inflicts the victim with a terminal fever. The person dies of the fever and is then reanimated as a walker. Say if someone’s hand was bitten, you could potentially save the bite victim’s life if you cut off their arm fast enough before the fever has a chance to set in. However, if you wait too long and allow the fever to set in, amputating the arm would be useless.
Hence why Reggie survived and Lee didn’t (if you choose to cut off Lee’s arm). Reggie’s arm was removed almost immediately after he was bitten while in the scenario with Lee by the time you have the option to cut off Lee’s arm it was too late.
