I doubt Eddie or Nate will appear because that asshole who hacked the game and leaked his findings spoiling it for everyone, didn't find any evidence of these 2 characters in the game files.
We'll probably meet Eddie as part of a new group while they are on the road to Wellington. Maybe Bonnie will know him from if Wyatt told her about him.
I really hope so! The interaction between him and Clem would be hilarious. Plus I'd love to see if he changed since we last saw him. If he's been able to survive this long, I'm sure he's seen some shit, plus the incident with Nate/Wyatt was probably super traumatizing. His appearance on the E4 title card was definitely intentional. Out of all the background characters on that slide, I don't think Eddie was put there just as filler. It would be a huge dick move on TTG's part if they put him them for shits and giggles and didn't include him in the game.

Out of all the remaining 400 DAYS cast who haven't appeared in S2 yet, I'd prefer Nate. Eddie's such an idiot (a likable idiot, but an idiot nonetheless) I just can't see him surviving two years into the zombie apocalypse.
